چکیده

Background: Opiate abuse has been critically increased in the world, especially Iran. Owing to association of opiate use with multiple human cancers and neurological disorders, seeking for genetic epigenetic effects opium can pave way early diagnosis major health defects addicted users. Accordingly, present study aimed determine methylation status promoter two genes, which are actively involved neurodevelopment cancer evolution. Methods: DNA was isolated from peripheral blood 28 abusers 19 healthy controls then subjected sonication. Sonicated DNAs undergone methylated immunoprecipitation-real time polymerase chain reaction (MeDIP-Real Time PCR) using specific primer pairs designed HOXA9 NISCH genes. Obtained data were analyzed SPSS software. Findings: genes found be significantly users compared (P<0.001) associated mean age regarding gene (P=0.002). Neither amount nor duration or route Conclusion: Hypermethylation as tumor suppressor opium-addicted individuals considered confirmatory evidence carcinogenesis opium. Further studies required figure out role alterations evolution among

Study of the role of siRNA mediated promoter methylation in DNMT3B knockdown and alteration of promoter methylation of CDH1, GSTP1 genes in MDA-MB -453 cell line

Promoter methylation is one of the main epigenetic mechanisms that lead to the inactivation of tumor suppressor genes during carcinogenesis. Due to the reversible nature of DNA methylation, many studies have been performed to correct theses epigenetic defects by inhibiting DNA methyltransferases (DNMTs). In this case novel therapeutics especially siRNA oligonucleotides have been used to specifi...

Two Steps Methylation Specific PCR for Assessment of APC Promoter Methylation in Gastric Adenocarcinoma

Gastric Cancer (GC) is the second most common cancer in the world and a leading cause of cancer-related mortality. Methylation of promoter CpG islands (CGIs) belonging to tumor suppressor genes causes transcriptional silencing of their corresponding genes leading to carcinogenesis and other disorders. Adenomatous Polyposis Coli (APC) a tumor suppressor gene is inactivated by methylation of prom...

Promoter Methylation of Four Tumor Suppressor Genes in Human Papillary Thyroid Carcinoma

Background & Objective: Papillary thyroid cancer (PTC) is considered to be the most common type of thyroid malignancies. Epigenetic alteration, in which the chromatin conformation and gene expression change without changing the sequence of DNA, can occur in some tumor suppressor genes and oncogenes. Methylation is the most common type of epigenetic alterations that can be an ex...
